pSP64TS

Excellent, simple, uncomplicated plasmid containing globin flanking sequences, a small polylinker and stop codons in all three ORF's. The polylinker was kept small due to the observation of Hemmati-Brivanlou and Harland that mRNA transcribed from plasmids with large polylinkers, such as Bluescript, only generate low levels of protein, possibly due to secondary structures in mRNA caused by the palindromic nature of the polylinkers. SpeI ends are compatible with XbaI and NheI, and BglII ends compatible with BamHI and Sau3AI. EcoRV is blunt. This plasmid is a derivative of pSP64T (Krieg and Melton, 1983). The modifications were made by Paul Wilson.


This sequence looks a bit strange, the BglII site directly precedes the EcoRV site.
