The gene cgl.2 encodes a secreted signaling protein belonging to the cerberus/gremlin family, which functions as a ligand for co-receptors including frizzled and lrp6. It acts as an antagonist of nodal, bmp4, and wnt8 signals during gastrulation. This antagonism is required for head formation by promoting anterior development and repressing posteriorizing signals. The protein is expressed in the dorsal marginal zone, specifically in the deep mesodermal cells of the dorsal lip of the blastopore and later in the prechordal plate and axial mesoderm.
