The dio3 gene encodes a type III iodothyronine deiodinase enzyme which is membrane-bound and inactivates thyroid hormones by catalyzing the outer ring deiodination of thyroxine (T4) and 3,5,3'-triiodothyronine (T3) to produce reverse T3 and 3,3'-diiodothyronine (T2), respectively. Its expression is detected in the brain and skin, playing a role in regulating local thyroid hormone concentrations during development, particularly controlling the timing of metamorphosis by modulating the availability of active T3 hormone.
