CMV promoter driving expression of of the epigenetic sensor, H3K9ac-mintbody (modification-specific intracellular antibody). Transgenic X. laevis ubiquitously expressing a fluorescent modification-specific intracellular antibody (Mintbody) that is able to track histone H3 lysine 9 acetylation (H3K9ac) in vivo through nuclear enhanced green fluorescent protein (EGFP) fluorescence. H3K9ac-Mintbody consists of a single-chain variable fragment (scFv) antibody against H3K9ac and enhanced Green Fluorescent Protein (eGFP). H3K9ac-Mintbody specifically binds to H3K9ac and consequently accumulates in nuclei. The dynamics of acetylation and active chromatin status can therefore be observed in vivo as intensity of nuclear EGFP fluorescence. |
