Description: CMV promoter driving expression of of the epigenetic sensor, H3K9ac-mintbody (modification-specific intracellular antibody). Transgenic X. laevis ubiquitously expressing a fluorescent modification-specific intracellular antibody (Mintbody) that is able to track histone H3 lysine 9 acetylation (H3K9ac) in vivo through nuclear enhanced green fluorescent protein (EGFP) fluorescence. H3K9ac-Mintbody consists of a single-chain variable fragment (scFv) antibody against H3K9ac and enhanced Green Fluorescent Protein (eGFP). H3K9ac-Mintbody specifically binds to H3K9ac and consequently accumulates in nuclei. The dynamics of acetylation and active chromatin status can therefore be observed in vivo as intensity of nuclear EGFP fluorescence.
Phenotype Description: Ubiquitous expression of green fluorescent sensor of histone H3 lysine 9 acetylation. Robust fluorescence observed in the nuclei of somites, and in regenerating notochord.
